Koch’s Postulates

Koch’s postulates are a set of criteria used to establish that a specific microorganism causes a specific disease.
Simple Explanation
To prove “this microbe causes this disease,” you should find it in the sick, isolate it pure, reproduce the disease in a host, and recover the same microbe again.

Formulated in the context of Robert Koch’s work on anthrax and tuberculosis, the classical postulates are:
- The microorganism is found in abundance in all organisms suffering from the disease, but not in healthy organisms.
- The microorganism can be isolated from a diseased organism and grown in pure culture.
- The cultured microorganism should cause disease when introduced into a healthy organism.
- The microorganism must be re-isolated from the inoculated, diseased experimental host and identified as identical to the original.
Modern practice recognizes exceptions: asymptomatic carriage, unculturable organisms, ethical limits on human challenge, opportunistic pathogens, toxins without ongoing infection, and polymicrobial disease. Molecular Koch’s postulates (Falkow and later frameworks) replace or supplement culture/challenge with evidence that a gene or factor is required for virulence.
Mechanism
Operational logic: association → isolation → causation via challenge → confirmation by re-isolation. Each step reduces alternative explanations (contamination, coincidence, secondary invaders).
