Microscopy
One-Sentence Definition
Microscopy is the direct visual examination of specimens at magnifications that reveal microorganisms, and it remains the fastest information a microbiology laboratory can produce.

Look at it. Within minutes you know whether there are bacteria, roughly what kind, and whether the specimen was worth processing.

Resolution limit of light microscopy is ~0.2 µm — which is why most viruses are invisible by light microscopy and why electron microscopy was required to see them.
Clinical Importance
- A Gram stain of CSF or a positive blood culture changes empirical therapy within minutes
- Specimen adequacy: sputum with many squamous epithelial cells is saliva, not sputum — microscopy rejects it before culture wastes two days
- Malaria diagnosis, ova and parasite examination, and fungal wet preps remain microscopy-dependent
Research Importance
- Single-cell dynamics, persister imaging, host–pathogen interaction imaging
Diagnostic Relevance
- Rapid, cheap, and universally available — but insensitive (roughly 10⁴–10⁵ organisms/mL needed for a smear) and operator-dependent

History
Antonie van Leeuwenhoek built the lenses that first revealed “animalcules” in the 1670s; microscopy is the founding technique of the discipline (Medical Microbiology History).
