Microscopy is the direct visual examination of specimens at magnifications that reveal microorganisms, and it remains the fastest information a microbiology laboratory can produce.

Resolution limit of light microscopy is ~0.2 µm — which is why most viruses are invisible by light microscopy and why electron microscopy was required to see them.

Clinical Importance

  • A Gram stain of CSF or a positive blood culture changes empirical therapy within minutes
  • Specimen adequacy: sputum with many squamous epithelial cells is saliva, not sputum — microscopy rejects it before culture wastes two days
  • Malaria diagnosis, ova and parasite examination, and fungal wet preps remain microscopy-dependent

Research Importance

  • Single-cell dynamics, persister imaging, host–pathogen interaction imaging

Diagnostic Relevance

  • Rapid, cheap, and universally available — but insensitive (roughly 10⁴–10⁵ organisms/mL needed for a smear) and operator-dependent

History
