Title Hitting the Wall: How a Beta-Lactam Actually Kills a Bacterium #shorts

Description Same drug, same door, different ending. In episode 1 a beta-lactamase destroyed ceftazidime in the periplasm. This time there is no enzyme: the drug reaches PBP1b, the enzyme that builds the peptidoglycan wall, and opens its beta-lactam ring on the active-site serine. The ring stays. Wall synthesis stops, the wall opens, and the inner membrane pushes through. Everything shown is a real structure: OmpF porin and E. coli LPS from a molecular dynamics simulation, PBP1b from PDB 3VMA, the bound drug pose from a cephalosporin acyl-enzyme (PDB 2C5W) placed on the conserved active-site motifs. The peptidoglycan mesh is stylized. Music: “Rain Over Kyoto Station” by The Mini Vandals (YouTube Audio Library).
